Pharmacological activity arises from agonism at the glucagon-like peptide-1 receptor, a G protein-coupled receptor expressed in the pancreas, the gastrointestinal tract, and the brainstem. Receptor activation raises intracellular cyclic adenosine monophosphate and enhances insulin release in a glucose-dependent manner, an effect that diminishes when blood glucose concentration is low. Other effects include slowed gastric emptying and hypothalamic satiety signalling. These pathways are described well. Receptor desensitisation rates across tissues, relative to the endogenous hormone, are still under investigation, and reported findings differ between laboratories.
The company that developed the compound filed it as a long-acting analogue, and it gained first approval in 2017 for type 2 diabetes. Later authorisations from several regulators extended the indication to chronic weight management, and the World Health Organization added the glucagon-like peptide-1 receptor agonist drug class to its model list of essential medicines in 2023. Production uses solid-phase peptide synthesis followed by side-chain conjugation and chromatographic purification. Supply constraints and cost differences across regions are well documented. Literature on long-term outcomes continues to grow, with many trials reporting surrogate endpoints rather than hard clinical endpoints.
The distinction between established facts and open questions matters here. That the peptide binds the GLP-1 receptor and stimulates insulin release in a glucose-dependent manner is well documented. How individual variability in receptor density, gastric emptying rate, and gut microbiome composition shapes response remains an active research area. Long-term outcomes beyond five years of continuous use are not yet fully characterized in published trials, and several extension studies are ongoing.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L cells after food intake. The molecule is a 31-amino-acid backbone modified at three positions to resist cleavage by dipeptidyl peptidase-4, the enzyme that degrades native GLP-1 within minutes. A lysine residue at position 26 carries a linker and a C18 fatty diacid, which promotes binding to serum albumin and slows renal clearance. These changes extend the circulating half-life from roughly two minutes to about one week in humans.
| Property | Value | Notes |
|---|---|---|
| Molecular class | 31-amino-acid peptide | Modified glucagon-like peptide-1 analogue |
| Molecular mass | approximately 4114 Da | Calculated for the free peptide backbone |
| Appearance | white to off-white powder | Typical of purified lyophilised peptide batches |
| Solubility class | freely soluble in water | Aqueous solubility improves at slightly alkaline pH |
| Typical storage | -20 degrees Celsius, dry, dark | Protect from repeated freeze-thaw cycles |
Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Lyophilized semaglutide powder is typically held at minus twenty degrees Celsius for long-term storage. At that temperature, solid-state degradation reactions proceed slowly and the peptide remains intact for extended periods. Repeated freeze-thaw cycles are best avoided because they promote aggregation and can shift the proportion of monomeric peptide present. Working aliquots are often prepared so that each portion is thawed only once, and desiccant is placed inside the vial to limit moisture uptake.
Serum protein binding dominates the pharmacokinetic profile. The attached chain associates strongly with albumin, shielding the peptide from enzymatic attack and slowing filtration by the kidney. This interaction extends the circulation half-life to roughly one week in humans, which supports weekly administration intervals. An oral version pairs the peptide with an absorption enhancer that transiently alters gastric epithelium, permitting limited uptake; bioavailability by that route is substantially lower than by injection.
Semaglutide belongs to the glucagon-like peptide-1 receptor agonist class, a group of synthetic peptides that imitate an incretin hormone released by intestinal L cells after food intake. Native GLP-1 circulates for only a few minutes because dipeptidyl peptidase-4 cleaves it rapidly. The hormone acts on pancreatic islets, the gastrointestinal tract, and several brain regions. Because the natural peptide is short-lived, development work concentrated on analogues that keep receptor activity while resisting enzymatic breakdown and renal clearance.
The semaglutide sequence is a 31-residue analogue of human GLP-1, altered at three positions relative to the parent hormone. Aminoisobutyric acid replaces alanine at position 8, arginine replaces lysine at position 34, and a lipophilic diacid is attached to lysine 26 through a short linker. These features are reported consistently in the structural literature. The position 8 substitution blocks recognition by dipeptidyl peptidase-4, while the attached chain drives strong, reversible association with a carrier protein in blood.
== Discovery == Studies conducted in the 1970s found that a series of N-formylmethionine-containing oligopeptides, including the most potent and best known member of this series, N-formylmethionine-leucyl-phenylalanine (fMLF or fMet-Leu-Phe), stimulated rabbit and human neutrophils by an apparent receptor-dependent mechanism to migrate in a directional pattern in classical laboratory assays of chemotaxis. Since these oligopeptides were produced by bacteria or synthetic analogs of such products, it was suggested that the N-formyl oligopeptides are important chemotactic factors and their receptors are important chemotactic factor receptors that act respectively as signaling and signal-recognizing elements to initiate inflammation responses in order to defend against bacterial invasion. Further studies defined a receptor for the N-formyl oligopeptides, formyl peptide receptor (FPR), so named based on its ability to bind and become activated by the oligopeptides. Two receptors where thereafter discovered and named FPR1 and FPR2 based on the similarity of their genes' predicted amino acid sequence to that of FPR rather than on any ability to bind or be activated by the formyl oligopeptides.

== Interactions == Von Willebrand Factor has been shown to interact with Collagen, type I, alpha 1. Recently, It has been reported that the cooperation and interactions within the von Willebrand Factors enhances the adsorption probability in the primary haemostasis. Such cooperation is proven by calculating the adsorption probability of flowing VWF once it crosses another adsorbed one. Such cooperation is held within a wide range of shear rates.
=== Impaired neutrophil migration and tumor necrosis factor- Alpha === Impaired neutrophil migration (caused by genetic defects, infections, and the aging process) causes increased macrophage activity that can lead to granuloma formation, a hallmark of NL. Furthermore, TNF-α is a pro-inflammatory cytokine that's activated by macrophages, and it plays a critical role in granuloma formation.
== Epidemiology == The first estimate of US prevalence for autoimmune diseases as a group was published in 1997 by Jacobson, et al. They reported US prevalence to be around 9 million, applying prevalence estimates for 24 diseases to a US population of 279 million. Jacobson's work was updated by Hayter & Cook in 2012. This study used Witebsky's postulates, as revised by Rose & Bona, to extend the list to 81 diseases and estimated overall cumulative US prevalence for the 81 autoimmune diseases at 5.0%, with 3.0% for males and 7.1% for females. In 2025, a study using electronic medical records from over 15 million patients at six large academic medical systems in the United States found a prevalence of 4.6% for autoimmune disease, based on a list of 105 conditions from The Rose and Mackay Textbook of Autoimmune Diseases. The study also found that over 34% of patients with an autoimmune disease had at least one other autoimmune condition, compared to cancer, where 8.1% of patients are diagnosed with a second primary malignancy.

== Other types == Other collagen types such as Collagen XX, which are smaller in size are mostly expressed in the connective tissue such as cartilage, tendon, and cornea while Collagen XIX is found in the skin, muscle cells, and the hippocampus. There are also collagens that are expressed in the basement membrane zones, which are extracellular matrices composed of macromolecule networks, and collagen XXII is one example located in the myotendinous junctions in the skeletal and heart muscles. COL22A1 is also included in this class.
Iridium-192 (symbol 192Ir) is a radioactive isotope of iridium, with a half-life of 73.82 days. It decays by emitting beta (β) particles and gamma (γ) radiation. 95.24% of 192Ir decays occur via β- emission, leading to 192Pt; the remaining 4.76% occur via electron capture to 192Os; both modes involve gamma emission. Iridium-192 is normally produced by neutron activation of natural-abundance iridium metal. Iridium-192 is a very strong gamma ray emitter, with a gamma dose constant of 1.54 μSv·h−1·MBq−1 at 30 cm, and a specific activity of 341 TBq·g−1 (9.22 kCi·g−1). There are seven principal gamma rays produced in its beta-minus decay, ranging from 296.0 to 612.5 keV, and two produced in its electron capture decay at 205.8 and 484.6 keV. It is commonly used as a gamma ray source in industrial radiography to locate flaws in metal components. It is also used in radiotherapy as a radiation source, in particular in brachytherapy. Iridium-192 has accounted for the majority of cases tracked by the U.S. Nuclear Regulatory Commission in which radioactive materials have gone missing in quantities large enough to make a dirty bomb. The metastable isomer 192m2Ir is iridium's most stable isomer. It decays solely by isomeric transition (to this ground state) with a half-life of 241 years, which is somewhat unusual for its long half-life and that said half-life greatly exceeds that of the ground state.
== Approval == Pramlintide has been approved on 3/16/2005 by the FDA, for use by type 1 and type 2 diabetic patients who use insulin.(subscription required) Pramlintide allows patients to use less insulin, lowers average blood sugar levels, and substantially reduces what otherwise would be a large unhealthy rise in blood sugar that occurs in diabetics right after eating. Apart from insulin analogs, pramlintide is the only drug approved by the FDA to lower blood sugar in type 1 diabetics since insulin in the early 1920s.
A cell-free system is an in vitro tool widely used to study biological reactions that happen within cells apart from a full cell system, thus reducing the complex interactions typically found when working in a whole cell. Subcellular fractions can be isolated by ultracentrifugation to provide molecular machinery that can be used in reactions in the absence of many of the other cellular components. Eukaryotic and prokaryotic cell internals have been used for creation of these simplified environments. These systems have enabled cell-free synthetic biology to emerge, providing control over what reaction is being examined, as well as its yield, and lessening the considerations otherwise invoked when working with more sensitive live cells.
While potassium chromate (K2CrO4) is used in the manufacture of a host of different commercial products such as inks, dyes, wood stains (by reacting with the tannic acid in wood), explosives, fireworks, fly paper, and safety matches, as well as in the tanning of leather, all of these uses are due to the chemistry of the chromate ion rather than to that of the potassium ion.

=== Sites for infusion sets === Infusion sets are most commonly placed on the abdomen, in a roughly semicircular area around and below the navel. Other sites include the upper leg, upper buttocks, hips, upper arms and lower back. Insulin absorption may vary from site to site, and therefore medical advice is required.
Even when the enzyme reaction does not result in a change in the absorbance of light, it can still be possible to use a spectrophotometric assay for the enzyme by using a coupled assay. Here, the product of one reaction is used as the substrate of another, easily detectable reaction. For example, figure 1 shows the coupled assay for the enzyme hexokinase, which can be assayed by coupling its production of glucose-6-phosphate to NADPH production, using glucose-6-phosphate dehydrogenase.
In order to obtain high yields of the desired target product, the reactants are continuously fed to a flow reactor and reacted with a more than two-fold excess of methylamine. In the downstream process steps, the excess methylamine and the water is removed and NMEA (bp. 160 °C) and MDEA (bp. 243 °C) are isolated from the product mixture by fractional distillation. The poly(methyl-ethanolamine) formed by further addition of ethylene oxide to methylethanolamine remains in the distillation bottoms.
=== Birth rate === According to one report, in 1924, shortly after the Free State's establishment, the new dominion had the "lowest birth-rate in the world". The report noted that amongst countries for which statistics were available (Ceylon, Chile, Japan, Spain, South Africa, the Netherlands, Canada, Germany, Australia, the United States, Britain, New Zealand, Finland, and the Irish Free State), Ceylon had the highest birth rate at 40.8 per 1,000 while the Irish Free State had a birth rate of just 18.6 per 1,000.
=== Models === A variety of theoretical frameworks exist to model optical, electronic, and structural properties of quantum dots. These may be broadly divided into quantum mechanical, semiclassical, and classical.

It is a glucagon-like peptide-1 receptor agonist, often grouped with the incretin mimetics. Its backbone is modified from the human hormone to resist enzymatic degradation and to bind albumin. These two features distinguish it from the native peptide.
No. It shares high sequence identity but carries non-natural modifications that alter its stability and clearance. The naturally occurring hormone is broken down within minutes, while the synthetic analogue circulates far longer.
Attachment to serum albumin reduces renal filtration of the peptide and shields it from enzymatic breakdown. The consequence is a much longer interval between administrations than unmodified peptides allow. The precise contribution of binding affinity to overall duration is still being quantified.
It is a modified version of the natural hormone, with three amino acid changes and a fatty acid side chain added. These edits extend its half-life from minutes to about one week. The core receptor activity is retained.
