Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.
GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Receptor activation occurs at GLP-1 receptors distributed across pancreatic islets, the hypothalamus, and the gastrointestinal tract. Binding triggers G protein signaling that raises cyclic AMP and enhances glucose-dependent insulin release. Because the effect depends on prevailing glucose levels, insulin secretion does not rise when blood sugar is already low. Signaling in the brain and gut also influences appetite and gastric emptying, which is why the compound appears in both metabolic and weight-related research literature.
Development began in the early 2010s with the goal of extending GLP-1 activity beyond the brief window achieved by native peptide infusion. The earliest approved formulation was a subcutaneous injection given once weekly. A later oral tablet pairs the peptide with an absorption enhancer, sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, usually shortened to SNAC. That carrier lowers local pH and helps the peptide cross gastric tissue. Both routes deliver the same active molecule.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, a gut hormone released after meals. Its backbone retains the GLP-1 sequence but incorporates two substitutions that slow enzymatic breakdown by dipeptidyl peptidase-4. A short polyethylene glycol linker and a C18 fatty diacid are attached to the peptide chain, allowing the molecule to bind serum albumin and remain in circulation far longer than the native hormone. The result is a circulating half-life measured in days rather than the minutes typical of endogenous GLP-1.

Material described as research-grade is not necessarily manufactured to pharmaceutical standards, and purity figures depend on the method used to obtain them. A certificate of analysis states the measured purity, the analytical technique, and the batch identifier, but the underlying data are not always included. Independent testing by a second laboratory is a common way to confirm identity and purity. Uncertainties remain about how storage history affects long-term stability, and about how well results from one laboratory transfer to another. Documentation of handling conditions supports comparison between batches.
Peptides are sensitive to temperature, light, oxygen, and repeated freeze-thaw cycles. Semaglutide in dry form is generally held at refrigerated temperatures, while reconstituted solutions require a defined short-term storage window. Vials should be kept in secondary packaging to limit photodegradation, and exposure to alkaline conditions is avoided because it accelerates chemical degradation. Adsorption to glass and some plastics can reduce the measured concentration of dilute solutions, so low-binding polypropylene containers are preferred for analytical work. Each transfer step introduces a small risk of contamination, and closed handling practices reduce that risk.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.
Quality control relies on pharmacopoeial monographs where they exist, combined with in-house specifications for identity, purity, water content, and counter-ion composition. Reference standards allow calibration across laboratories, although certified materials for every analogue are not universally obtainable. Batch records, chromatograms, and mass spectra form the documentation trail. Regulatory classification varies by jurisdiction and intended use, and research-grade material differs from pharmaceutical-grade material in testing scope. Analytical uncertainty is often expressed as relative standard deviation across replicate injections.
Lyophilised semaglutide is generally held at -20 °C or below, protected from light and moisture. Reconstituted solutions are typically kept at 2-8 °C and used within a defined window because degradation accumulates over time. Repeated freeze-thaw cycles are discouraged, since each cycle can promote aggregation and reduce monomeric content. Room-temperature stability of the solid has been examined in some studies but remains incompletely characterised for long durations, so cold storage is the conservative default for research material.
A simplified explanation for the integration of the pressure over the contact area may be stated as follows: Consider a cube immersed in a fluid with the upper surface horizontal. The sides are identical in area, and have the same depth distribution, therefore they also have the same pressure distribution, and consequently the same total force resulting from hydrostatic pressure, exerted perpendicular to the plane of the surface of each side. There are two pairs of opposing sides, therefore the resultant horizontal forces balance in both orthogonal directions, and the resultant force is zero. The upward force on the cube is the pressure on the bottom surface integrated over its area. The surface is at constant depth, so the pressure is constant. Therefore, the integral of the pressure over the area of the horizontal bottom surface of the cube is the hydrostatic pressure at that depth multiplied by the area of the bottom surface. Similarly, the downward force on the cube is the pressure on the top surface integrated over its area. The surface is at constant depth, so the pressure is constant. Therefore, the integral of the pressure over the area of the horizontal top surface of the cube is the hydrostatic pressure at that depth multiplied by the area of the top surface.
Dried spirulina is 5% water, 24% carbohydrates, 8% fat, and 57% protein (table). In a reference amount of 100 g (3.5 oz), dried spirulina powder supplies 290 kilocalories (1,200 kJ) and is a rich source (20% or more of the Daily Value, DV) of numerous essential nutrients, particularly B vitamins (thiamin, riboflavin, and niacin), and dietary minerals, such as iron and manganese (table). The lipid content of spirulina is about 8% by weight. The polyunsaturated fatty acids include gamma-linolenic acid and linoleic acid. In contrast to the "high" content reported in a 2003 study, two other analyses found low levels of omega-3 fatty acids in spirulina.
The first injector pen, branded "Penject", was introduced in 1983 to administer insulin products, following trials begun in 1981 at Southern General Hospital, Scotland, initiated by Sheila Reith who has conceived the device, and funded by Diabetes UK. The invention was announced, and the preliminary trial described, in a January 1981 paper in The Lancet. The patent rights were later sold to Novo Nordisk by Greater Glasgow Health Board. After their introduction, insulin pens had a slow adoption in the United States, with only 2% of insulin being injected via pen in 1999. A major barrier to adoption in the United States was the increased up-front cost of insulin pens compared to traditional injections. Pen adoption in the United States accelerated after studies showed that the higher up-front cost of insulin pens was offset by the increase in compliance, which decreased overall healthcare costs. Historically, pen needles were manufactured in lengths up to 12.7mm. Over time, pen needles designed for insulin pens have become shorter, and a 4mm long needle is considered sufficient for most people to administer subcutaneously correctly. In 1989, an injector pen form of human growth hormone was licensed in New Zealand.
The chemical structure of penicillin was first suggested by Abraham in 1942. Dorothy Hodgkin determined the correct chemical structure of penicillin using X-ray crystallography at Oxford in 1945. In 1945, the US Committee on Medical Research and the British Medical Research Council jointly published in Science chemical analyses conducted at different universities, pharmaceutical companies and government research departments. The report announced the existence of different forms of penicillin compounds which all shared the same structural component called β-lactam. The penicillins were designated by Roman numerals in UK (penicillin I, II, III and IV) in order of their discoveries, and known by letters (F, G, X, and K) referring to their origins or sources in the US, as below:

The primary purpose of a tablet counter (also known as a pill counter or drug counter) is to accurately count prescription medications in tablet or capsule form to aid the requirement for patient medication safety, to increase efficiency and reduce costs for the typical pharmacy. Newer versions of this counting device include advanced software to continue to improve safety for the patient who is receiving the prescription, ensuring that the pharmacy staff dispense the right medication at correct dosage strength for the right patient. (see also medication safety). Today's pharmacy industry recognizes the need for heightened vigilance against medication errors across the entire spectrum. A wealth of research has been conducted regarding the prevalence of medication errors and the ability of technology to decrease or eliminate such errors. (See the March 2003 landmark study by Auburn University's Center for Pharmacy Operations and Designs). Prescription dispensing safety and accuracy in the pharmacy are an essential part of ensuring the right patient gets the right medication at the right dosage. A trend in pharmacy is to place a greater reliance on technology and pharmacy automation to minimize the chance of human error and speed up the process of dispensing. Pharmacy management generally sees technology as a solution to industry challenges like staffing shortages, prescription volume increases, long and hectic work hours and complicated insurance reimbursement procedures.
==== Conclusions ==== As the cathode rays carry a charge of negative electricity, are deflected by an electrostatic force as if they were negatively electrified, and are acted on by a magnetic force in just the way in which this force would act on a negatively electrified body moving along the path of these rays, I can see no escape from the conclusion that they are charges of negative electricity carried by particles of matter. As to the source of these particles, Thomson believed they emerged from the molecules of gas in the vicinity of the cathode.
== History == MrMed was conceived in August 2020 when Devashish Singh and Saurab Jain noticed challenges in specialty medicines. Despite facing initial setbacks due to the COVID Delta wave in India, which prompted the founders and their team to prioritize community assistance, MrMed was officially launched in June 2021. The company launched its mobile app in March 2023 for access to speciality medicines for conditions such as cancer, transplants, kidney, or liver diseases. It also raised $500,000 in funding from the Tamil Nadu Emerging Sector Seed Fund (TNESSF) and angel investors. On 8 December 2025, the company announced a strategic partnership with the American multinational pharmaceutical company Eli Lilly to support the distribution of specialty medicines in India.
== Safe handling == Proper handling of affected or damaged materials prevents mold from spreading. Because mold spores are released into the air, it is recommended to wear an NIOSH-rated respirator before coming into contact with affected materials. Nitrile gloves should be worn to protect the skin from contact with mold. In extreme cases, a full-body hazmat suit may be required. Materials must always be treated on a surface that can be cleaned with bleach or on neutral, disposable materials such as unprinted newsprint.

=== Nationality and travel history === Many nations implement border controls restricting the entry of people of certain nationalities or who have visited certain countries. For instance, Georgia refuses entry to holders of passports issued by the Republic of China. Similarly, since April 2017, nationals of Bangladesh, Pakistan, Sudan, Syria, Yemen, and Iran have been banned from entering the parts of eastern Libya under the control of the Tobruk government. The majority of Arab countries, as well as Iran and Malaysia, ban Israeli citizens, however exceptional entry to Malaysia is possible with approval from the Ministry of Home Affairs. Certain countries may also restrict entry to those with Israeli stamps or visas in their passports. As a result of tension over the Artsakh dispute, Azerbaijan currently forbids entry to Armenian citizens as well as to individuals with proof of travel to Artsakh. Between September 2017 and January 2021, the United States did not issue new visas to nationals of Iran, North Korea, Libya, Somalia, Syria, or Yemen pursuant to restrictions imposed by the Trump administration, which were subsequently repealed by the Biden administration on 20 January 2021. While in force, the restrictions were conditional and could be lifted if the countries affected meet the required security standards specified by the Trump administration, and dual citizens of these countries could still enter if they presented a passport from a non-designated country.
polyploid (of a cell or organism) Having more than two homologous copies of each chromosome; i.e. any ploidy level that is greater than diploid. Polyploidy may occur as a normal condition of chromosomes in certain cells or even entire organisms, or it may result from errors in cell division or mutations causing the duplication of the entire chromosome set.
According to his children John and Priscilla Tolkien: In later years, he would occasionally talk of being at the front: of the horrors of the first German gas attack, of the utter exhaustion and ominous quiet after a bombardment, of the whining scream of the shells, and the endless marching, always on foot, through a devastated landscape, sometimes carrying the men's equipment as well as his own to encourage them to keep going. ... Some remarkable relics survive from that time: a trench map he drew himself; pencil-written orders to carry bombs to the 'fighting line'. Many of his dearest school friends were killed in the war. Among their number were Rob Gilson of the Tea Club and Barrovian Society, who was killed on the first day of the Somme while leading his men in the assault on Beaumont Hamel. Fellow T.C.B.S. member Geoffrey Smith was killed during the battle, when a German artillery shell landed on a first-aid post. Tolkien's battalion was almost completely wiped out following his return to England.
==== Suppression of Cossack autonomy in the Russian Empire ==== Historian Gary Dean Peterson writes: "With all this unrest, Ivan Mazepa of the Ukrainian Cossacks was looking for an opportunity to secure independence from Russia and Poland". In response to Mazepa's alliance with Charles XII of Sweden, Peter I ordered the sacking of the then capital of the Hetmanate, Baturyn. The city was burnt and looted, and 11,000 to 14,000 of its inhabitants were killed. The destruction of the Hetmanate's capital was a signal to Mazepa and the Hetmanate's inhabitants of severe punishment for disloyalty to the Tsar's authority. The Zaporizhian Sich at Chortomlyk, which had existed since 1652, was also destroyed by Peter I's forces in 1709, in retribution for decision of its otaman Kost Hordiyenko, to ally with Mazepa. Under Russian rule, the Cossack nation of the Zaporozhian Host was divided into two autonomous republics of the Russian Tsardom: the Cossack Hetmanate, and the more independent Zaporizhia. These organizations gradually lost their autonomy, and were abolished by Catherine II in the late 18th century. The Hetmanate became the governorship of Little Russia, and Zaporizhia was absorbed into New Russia.

It is given either as a once-weekly subcutaneous injection or as an oral tablet taken once daily. The two forms use different absorption strategies, so they are not interchangeable on a milligram-for-milligram basis.
Structural modifications, including a fatty acid side chain and non-natural amino acid substitutions, slow enzymatic breakdown and promote albumin binding. These changes support once-weekly dosing rather than twice-daily administration.
The pathways involving insulin, glucagon, gastric emptying, and appetite signaling are well described. How much each pathway contributes to weight reduction in a given person is not fully established.
Native GLP-1 is degraded within minutes by circulating enzymes. The synthetic version carries substitutions at positions that block enzymatic cleavage, plus a fatty acid side chain that promotes albumin binding. These two changes together extend circulation time from minutes to roughly a week.
