Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.
GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 nm is the standard purity method, reported as area percent. Mass spectrometry, usually with electrospray ionisation, confirms identity and reveals covalent modifications. Size-exclusion chromatography quantifies aggregates and fragments. Peptide mapping after enzymatic digestion localises changes to specific sequence regions. Circular dichroism and infrared spectroscopy report on secondary structure, while light scattering tracks particle formation in liquid formulations. No single technique captures every quality attribute.
Quality control relies on pharmacopoeial monographs where they exist, combined with in-house specifications for identity, purity, water content, and counter-ion composition. Reference standards allow calibration across laboratories, although certified materials for every analogue are not universally obtainable. Batch records, chromatograms, and mass spectra form the documentation trail. Regulatory classification varies by jurisdiction and intended use, and research-grade material differs from pharmaceutical-grade material in testing scope. Analytical uncertainty is often expressed as relative standard deviation across replicate injections.

Stability studies focus on deamidation of asparagine and glutamine residues, oxidation of methionine, and aggregation into higher-order species. The fatty acid side chain adds susceptibility to oxidative change and can promote self-association at high concentration. Lyophilised material is comparatively robust when kept cold and dry, while aqueous solutions require refrigeration and protection from light. Forced degradation experiments under heat, acid, base, and peroxide conditions establish the specificity of each analytical method. Which degradation route dominates under real storage conditions depends on the formulation and stays formulation-specific.
Handling guidance for research quantities calls for single-use aliquots, an inert atmosphere where practical, and avoidance of repeated freeze-thaw cycles that accelerate aggregation. Certificates of analysis typically report purity by peak area, water content, counter-ion identity, and residual solvent levels. In the scientific literature the compound is usually described by its full amino acid sequence, its registry number, or its structural class rather than by any proprietary label. Reporting standards vary between journals, and reviewers increasingly request raw chromatograms alongside tabulated purity figures. Whether current purity thresholds are adequate for every experimental context is debated.
Reversed-phase high-performance liquid chromatography with ultraviolet detection is the dominant approach for peptide purity assessment, usually paired with mass spectrometry to confirm molecular mass and sequence. Peptide mapping by enzymatic digestion and tandem mass spectrometry locates modifications such as deamidation and oxidation. Quantitation in plasma matrices can be performed by LC-MS/MS after solid-phase extraction. Method validation follows general guidance on accuracy, precision, linearity, and limits of detection. Comparability of results between laboratories, when no shared reference standard is available, remains an open question.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, a gut hormone released after meals. Its backbone retains the GLP-1 sequence but incorporates two substitutions that slow enzymatic breakdown by dipeptidyl peptidase-4. A short polyethylene glycol linker and a C18 fatty diacid are attached to the peptide chain, allowing the molecule to bind serum albumin and remain in circulation far longer than the native hormone. The result is a circulating half-life measured in days rather than the minutes typical of endogenous GLP-1.
Receptor activation occurs at GLP-1 receptors distributed across pancreatic islets, the hypothalamus, and the gastrointestinal tract. Binding triggers G protein signaling that raises cyclic AMP and enhances glucose-dependent insulin release. Because the effect depends on prevailing glucose levels, insulin secretion does not rise when blood sugar is already low. Signaling in the brain and gut also influences appetite and gastric emptying, which is why the compound appears in both metabolic and weight-related research literature.
Striated muscle tissue is a muscle tissue that features repeating functional units called sarcomeres. Under the microscope, sarcomeres are visible along muscle fibers, giving a striated appearance to the tissue. The two types of striated muscle are skeletal muscle and cardiac muscle.
In the 1930s and later, epidemiologists from Japan, the UK, and the US, including Richard Doll and various others, reported greater rates of death from lung cancer following occupational exposure to PAH-rich environments among workers in coke ovens and coal carbonization and gasification processes.
Protesters chanted slogans against "federalism" and demanded "full independence" for Syria, with slogans praising Hikmat al-Hijri. On 23 August, the National Guard was formed, with the aim of consolidating military efforts to confront what it described as "Salafist-jihadist gangs", with large military groups such as Anti-Terrorism Force, Al-Jabal Brigade, Sheikh al-Karama Forces, the As-Suwayda Operations Room, Al-Fahd Forces, with small groups, and the Jaysh al-Muwahhidin included. On 25 August, according to Enab Baladi, other groups that later joined include "Men of Dignity, the Unitarian Popular Resistance, Southern Shield Forces, Mountain Shield Forces, Lions of the Mountain Forces, Shield of Tawhid, Sahwat al-Khedr Shield, Mimas and Tal al-Lawz, Men of al-Lajat and Men of al-Kafr", while 164th Brigade joined on 24 August, and the Suwayda Military Council joined on 27 August.
=== Location === The company moved from 3200 Hillview Ave to the former headquarters of Facebook at 1601 S. California Ave in June 2012. By 2017 Theranos was headquartered at 1701 Page Mill Road, Palo Alto, California, paying a $1 million per month lease for the Stanford Research Park building. It had laboratories in Newark, California, and Scottsdale, Arizona. In 2017, when Theranos was still denying its publicly reported fraud, the company vacated its Page Mill headquarters, moving remaining staff into the Newark, California, laboratories; Stanford University School of Medicine converted the Page Mill building to offices and medical laboratories.
CPA can be taken by mouth or by injection into muscle. It has near-complete oral bioavailability, is highly and exclusively bound to albumin in terms of plasma protein binding, is metabolized in the liver by hydroxylation and conjugation, has 15β-hydroxycyproterone acetate (15β-OH-CPA) as a single major active metabolite, has a long elimination half-life of about 2 to 4 days regardless of route of administration, and is excreted in feces primarily and to a lesser extent in urine.

== Importance == D. dadantii has been associated with bacterial soft rot diseases of a majority of foliage plants, numerous flowering plants and many vegetables. It is a major pathogen for many economic crops such as potatoes, banana and pineapple in addition to ornamental house plants. It causes blackleg of potato. In addition to the pathogen having important negative consequences, D. dadantii is being used for its positive contributions. Most noble of its contributions is an enzyme, asparaginase, being used in conjunction with other chemotherapeutic agents for treatment of acute lymphoblastic leukemia (ALL) and non-Hodgkin's lymphoma in patients who have had allergic reactions to E. coli derived asparaginase Elspar or pegaspargase (Oncaspar). Secondly, with a strong governmental push towards increasing renewable fuel resources, D. dadantii is being studied for its utilization in ethanol fuel production and its ability to ferment and break down cell walls and pectins as an alternative to E. coli. Although not as effective as D. dadantii, some genes from D. dadantii were added to E. coli through genetic engineering to allow for pectin degradation by E. coli.
=== Other methods === Burning sulfur together with saltpeter (potassium nitrate, KNO3), in the presence of steam, has been used historically. As saltpeter decomposes, it oxidizes the sulfur to SO3, which combines with water to produce sulfuric acid. Prior to 1900, most sulfuric acid was manufactured by the lead chamber process. As late as 1940, up to 50% of sulfuric acid manufactured in the United States was produced by chamber process plants. A wide variety of laboratory syntheses are known, and typically begin from sulfur dioxide or an equivalent salt. In the metabisulfite method, hydrochloric acid reacts with metabisulfite to produce sulfur dioxide vapors. The gas is bubbled through nitric acid, which will release brown/red vapors of nitrogen dioxide as the reaction proceeds. The completion of the reaction is indicated by the ceasing of the fumes. This method conveniently does not produce an inseparable mist.
The International Rice Research Institute and the International Maize and Wheat Improvement Center are part of a consortium of agricultural research organizations known as CGIAR. Costing around $600 million, over 50 years, the revolution brought new farming technology, increased productivity, expanded crop yields and mass fertilization to many countries throughout the world. Later it funded over $100 million of plant biotechnology research and trained over four hundred scientists from Asia, Africa and Latin America. It also invested in the production of transgenic crops, including rice and maize. In 1999, the then president Gordon Conway addressed the Monsanto Company board of directors, warning of the possible social and environmental dangers of this biotechnology, and requesting them to disavow the use of so-called terminator genes; the company later complied. In the 1990s, the foundation shifted its agriculture work and emphasis to Africa; in 2006, it joined with the Bill & Melinda Gates Foundation in a $150 million effort to fight hunger in the continent through improved agricultural productivity. In an interview marking the 100 year anniversary of the Rockefeller Foundation, Judith Rodin explained to This Is Africa that Rockefeller has been involved in Africa since their beginning in three main areas – health, agriculture and education, though agriculture has been and continues to be their largest investment in Africa.
== Function == The primary function of DPEP1 is to hydrolyze various dipeptides in renal metabolism. Specifically, it has been found to hydrolyze glutathione and its conjugates such as leukotriene D (Kozak and Tate, 1982). Several pieces of evidence suggest that DPEP1 is also responsible for the hydrolysis of the beta-lactam ring of various THM-class antibiotics, such as penem and carbapenem (Campbell et al., 1984). First, the metabolism of these THM-class antibiotics is known to be localized in the kidney, specifically by a membrane-bound protein. Second, the metabolism of these antibiotics is significantly hindered when the zinc concentration is altered, suggesting the enzyme responsible for the drugs’ metabolism is a zinc-metalloenzyme. Finally, when DPEP1 was experimentally added to penem and carbapenem antibiotics in vitro, the resulting products were structurally identical to their respective metabolites found in an organism's urine (8). The hydrolysis of these antibiotics hinders their antibacterial capabilities, so information on the specific structure of DPEPI is highly sought after in order to find viable inhibitors that could be taken along with these antibiotics to make them more effective. Earlier, beta-lactamase enzymes were thought to occur only in bacteria, where their probable function was in protecting the organisms against the action of beta-lactam antibiotics. These antibiotics exhibit selective toxicity against bacteria but virtual inertness against many eukaryotic cells (Adachi et al., 1990).[supplied by OMIM]

Accounts of exactly what happened vary but it seems likely that he was mockingly offering it to the Labour benches, not, as some alleged, "brandishing" it – an illusion caused by Prior pulling his other arm down. Thatcher was furious. Speaker Thomas suspended the sitting and made Heseltine wait until next day to apologise so that tempers could cool. Heseltine was faced with calls for his resignation from the Shadow Cabinet; he thought it would play well with the public, but in Crick's view it helped to cement a reputation for impulsiveness and poor judgement. In autumn 1976 Heseltine was reshuffled, against his will, to the job of Shadow Environment Secretary. He was particularly cross at having to give up the job of Shadow Industry Secretary to John Biffen. He accepted on condition that he would not have to take the Environment job when the Conservatives returned to office. As Benn had given way to Eric Varley there was no longer such a need for aggressive campaigning on Industry, and Thatcher, who had herself been Shadow Environment Secretary in 1974, wanted him to campaign on council house sales (Heseltine offered up to 50% discounts for tenants who bought their homes) and reform of the rates, as she thought his predecessor Timothy Raison ineffective.

It is given either as a once-weekly subcutaneous injection or as an oral tablet taken once daily. The two forms use different absorption strategies, so they are not interchangeable on a milligram-for-milligram basis.
Structural modifications, including a fatty acid side chain and non-natural amino acid substitutions, slow enzymatic breakdown and promote albumin binding. These changes support once-weekly dosing rather than twice-daily administration.
The pathways involving insulin, glucagon, gastric emptying, and appetite signaling are well described. How much each pathway contributes to weight reduction in a given person is not fully established.
Reverse-phase high-performance liquid chromatography with ultraviolet detection is the usual choice, with results reported as area percent. Complementary methods such as size-exclusion chromatography and mass spectrometry are needed because a single separation cannot resolve every impurity class. Purity figures are therefore method dependent and should always be read alongside the technique used.
