== Mechanism == Antibodies against voltage-gated potassium channels (VGKC), which are detectable in about 40% of patients with acquired neuromytonia, have been implicated in Morvan's pathophysiology. Raised serum levels of antibodies to VGKCs have been reported in three patients with Morvan's Syndrome. Binding of serum from a patient with Morvan's Syndrome to the hippocampus in a similar pattern of antibodies to known VGKC suggest that these antibodies can also cause CNS dysfunction. Additional antibodies against neuromuscular junction channels and receptors have also been described. Experimental evidence exists that these anti-VGKC antibodies cause nerve hyperexcitability by suppression of voltage gated K+ outward currents, whereas other, yet undefined humoral factors have been implicated in anti-VGKC antibody negative neuromyotonia. It is believed that antibodies to the Shaker-type K+ channels (the Kv1 family) are the type of potassium channel most strongly associated with acquired neuromyotonia and Morvan's Syndrome. Whether VGKC antibodies play a pathogenic role in the encephalopathy as they do in the peripheral nervous system is as yet unclear. It has been suggested that the VGKC antibodies may cross the blood–brain barrier and act centrally, binding predominantly to thalamic and striatal neurons causing encephalopathic and autonomic features.

== Limitations == Limitations of magnetic drug delivery can range from their inherent magnetic properties to interactions with bodily barriers. When magnetic nanoparticles are in the bloodstream, they have high solubility and ionic strength, allowing them to interact with plasma proteins, stimulating the immune system to further inhibit their function. Additionally, the proportion of the nanoparticle size to the target tissue has shown limitations in effective drug delivery, especially in the kidneys and the brain. Intracellular barriers include the removal of the magnetic nanoparticles from the target membrane by ligand-dependent endocytosis followed by separation via acidification in the endosome chamber. Other barriers to consider are the depth of the target tissue, vascular sources, body weight, the speed and amount of blood flow to the target tissue, distance from the field source, injection route, and tumor volume. However, the use of magnetic nanoparticles is more effective when used in near-surface tissues that have slower blood flow, allowing for diffusion and/or endocytosis of nanoparticles into the tissue. Another limitation involves the accumulation of nanoparticles only 5 mm away from an external magnet. An accumulation distance of 5 mm may not be sufficient in larger applications of magnetic drug delivery. This may be effective enough for sites in closer proximity to the surface of the body, but when the site of interest is deeper within tissue, then the advantage of using magnetic nanoparticles for delivery decreases exponentially.
== Mechanism == According to later research on its mechanism of action, α-bungarotoxin binds irreversibly to the postsynaptic nicotinic acetylcholine receptor (nAChR) at the neuromuscular junction. It inhibits the action of acetylcholine competitively, leading to respiratory failure, paralysis and even death. α-bungarotoxin specifically targets the nervous system, interfering with the nerve impulse transmission. The primary target of the toxin is the neuromuscular junction of skeletal muscles, where the motor nerve terminal and the nicotinic acetylcholine receptor are the major target sites. Their neurotoxic effect is often referred to as resistant neurotoxicity. This is because of the damage caused to nerve terminals that leads to acetylcholine depletion at the neuromuscular junction. The regeneration of the synapses can take days, which prolongs the paralysis and recovery process for the victim. In addition, the severity of the paralysis ranges from mild to life-threatening depending on the degree of envenomation, its composition and the early therapeutic intervention.
